dc.description.abstract: Проанализированы литературные и собственные данные по морфологии четырех видов скребней от черноморских рыб, относившихся ранее к роду Acanthocephaloides (А. propinquus (Dujardin, 1845) Meyer, 1933; A. incrassatus (Molin, 1858) Meyer, 1933; A. kostylewi Meyer, 1933, A. rhytidotes (Porta, 1905) Belofastova et Kornijchuk, 2000). Установлено: A. incrassatus в Черном море отсутствует; A. propinquus — широкоспецифичный вид с 4-6 крючьями в 10-12 рядах на хоботке, паразитирует в кишечнике у 29 видов черноморских рыб; узкоспецифичный вид A. kostylewi от морского языка Solea nasutus (Pallas, 1814) с 5-6 крючьями в 12-14 рядах на хоботке морфологически близок как к Solearhynchus soleae (Porta, 1905) Buron et Maillard, 1985, так и к A. rhytidotes (Monticelli, 1905). Все 3 видовых названия, очевидно, являются синонимами. Приоритетность какого-либо из них остается спорной.

dc.description.abstract: В статье приведены сведения о специализированной рабочей коллекции каротиногенных микроводорослей отдела физиологии животных и биохимии Федерального исследовательского центра «Институт биологии южных морей имени А. О. Ковалевского РАН» (ФИЦ ИнБЮМ), созданной в рамках научной и прикладной тематик института для исследования механизмов стресс-толерантности у эврибионтных и экстремофильных одноклеточных фототрофов, а также для выявления коммерчески значимых источников высокоценных в медицинском и пищевом отношении кетокаротиноидов группы астаксантина. Коллекция насчитывает 44 штамма микроводорослей различной таксономической и экологической специализации с выраженной способностью к гиперсинтезу вторичных каротиноидов и липидов при экстремальных внешних воздействиях (высыхание, острое голодание, высокая освещённость, температура и солёность, действие токсикантов и др.). Основными способами пополнения фонда являются направленный обмен каротиногенными видами с ведущими российскими и зарубежными коллекциями микроводорослей и собственные полевые сборы в причерноморских зонах Крыма и Кавказа. Большинство штаммов в коллекции — представители двух порядков класса Chlorophyceae [Chlamydomonadales (25 штаммов) и Sphaeropleales (15 штаммов)], так как именно в этих порядках явление вторичного каротиногенеза распространено наиболее широко. Среди них преобладают обитатели эфемерных пресноводных водоёмов, аэрофильные и почвенные микроводоросли. Все штаммы поддерживаются в состоянии альгологически чистых культур при контролируемых условиях на агаризованных минеральных средах. Описания вариететов коллекции включают следующие сведения: а) современный таксономический статус вида, верифицированный с учётом обновлённых данных депонирующих коллекций и альгологических баз AlgaeBase и NCBI Taxonomy Browser; б) базионим и известные синонимы вида; в) время и источник поступления штамма в коллекцию; г) фамилию автора, географическое место и биотоп, из которого штамм был изолирован; д) номер штамма в NCBI (если есть); е) питательную среду, на которой штамм поддерживается в коллекции ФИЦ ИнБЮМ. Проанализировано значение коллекции для проведения морфобиологических и физиолого-биохимических исследований особенностей роста, вторичного каротиногенеза и биотехнологического потенциала зелёных микроводорослей.
dc.description: The article contains information on the specialized working collection of carotenogenic microalgae maintained by the staff of the animal physiology and biochemistry department of A. O. Kovalevsky Institute of Biology of the Southern Seas of RAS (IBSS). The collection was established within the framework of IBSS scientific and applied research to study the mechanisms of stress tolerance in eurybiontic and extremophilic single-celled phototrophs and to identify commercially significant sources of highly valuable ketocarotenoids of astaxanthin group used for medicine and food production. The collection contains 44 microalgal strains of various taxonomic and ecological specialization with a pronounced ability to hypersynthesize secondary carotenoids and lipids under extreme conditions (drying, nutrient starvation, high-intensity illumination, extreme temperature and salinity, effect of toxicants, etc.). The main ways to replenish the fund are direct exchange of carotenogenic species with leading Russian and foreign collections of microalgae and own field sampling in the Black Sea areas of Crimea and Caucasus. The majority of strains in the collection represent two orders of the class Chlorophyceae: Chlamydomonadales (25 strains) and Sphaeropleales (15 strains), since the phenomenon of secondary carotenogenesis is widespread in these orders. Out of them, inhabitants of ephemeral freshwater ponds predominate, as well as aerophilic and soil microalgae. All strains are maintained under controlled conditions on agarized mineral media as pure cultures. Description of the collection accession includes the following data: a) current taxonomic status of the species verified according to updated information from corresponding collections and algological databases, namely AlgaeBase and NCBI Taxonomy Browser; b) species basionym and known synonyms; c) date and source of the strain deposition; d) author’s surname, geographic location, and biotope, from which the strain was isolated; e) accession number of sequences associated with the strain in NCBI (if any); and f) nutrient medium, on which the strain is maintained in the IBSS collection. The significance of the collection for morphological, biological, physiological, and biochemical studies of growth, secondary carotenogenesis, and biotechnological potential in green microalgae is discussed.

dc.description.abstract: Надёжное сохранение культур микроводорослей и создание генетических банков штаммов — одна из важных задач современной биологии. В каталоге Всемирной федерации культур в базе WDCM CCINFO на сегодняшний день зарегистрировано 792 коллекции различных культивируемых организмов из 76 стран. Это самая обширная сводная база данных, включающая как известные крупные коллекции, так и небольшие хранилища исследовательских и образовательных учреждений со всего мира. В базе представлено 47 альгологических коллекций и 80 коллекций микроорганизмов, которые также включают культуры микроводорослей и цианобактерий. В России зарегистрировано всего 30 биологических коллекций; фонды только 13 из них включают штаммы водорослей. Самый распространённый способ хранения культур микроводорослей — метод их периодических пересевов на жидкие среды или агар. Его используют в 127 коллекциях (99 % от общего количества в каталоге). Также применяют криоконсервацию — в 33 коллекциях (27 %), лиофилизацию — в 13 (11 %), L-высушивание — в 5 (4 %), замораживание — в 19 (16 %), иммобилизацию в альгинатных бусинках — в 1 (0,8 %). Между тем при использовании этих методов изменяются морфологические и функциональные свойства клеток сохраняемых культур и происходит их измельчание. Кроме того, поддержание культур в жизнеспособном состоянии трудоёмко и требует дорогостоящего оборудования. При этом хранение микроводорослей, переведённых в состояние ангидробиоза путём их обезвоживания, просто и экономически выгодно. Ангидробиоз — глубокое и длительное торможение метаболизма, обратимое при благоприятных условиях; это достаточно распространённое явление в природе. Единственная коллекция из базы WDCM CCINFO, для которой применяют способ перевода клеток в покоящееся состояние путём ангидробиоза (для почвенных водорослей) — коллекция культур водорослей Киевского национального университета (ACKU WDCM 994). Многолетние опыты по переводу микроводорослей в состояние ангидробиоза позволили разработать метод их длительного хранения без использования питательных сред, включающий перевод клеток в состояние ангидробиоза, их сохранение в дегидратированном состоянии и последующее выведение в активную культуру. С целью поддержания альгологического биоразнообразия на базе ФИЦ ИнБЮМ создано хранилище микроводорослей, переведённых в состояние ангидробиоза; их при необходимости можно вывести в активные культуры. Объектами стали морские одноклеточные водоросли, а также пресноводные и галобные виды низших фототрофов, перспективные для аквакультуры и биотехнологии. Культуры получены в виде инокулята из коллекции живых культур планктонных микроводорослей ФИЦ ИнБЮМ. Водоросли выращивали в накопительном режиме при постоянном освещении. Биомассу собирали во время культивирования альгологически чистых культур микроводорослей на стадии замедления роста или на стационарной стадии. Клетки отделяли от культуральной среды центрифугированием или путём их фильтрации на планктонном сите. Затем водоросли обезвоживали и хранили в герметичных зиплок-пакетах, помещённых в пластиковые ёмкости объёмом от 100 до 500 мл, при температуре +18...+21 °C в темноте в специально оборудованном помещении. Основная часть коллекции представлена штаммами из отделов Chlorophyta, Cyanophyta, Bacillariophyta, Rodophyta. В статье приведены список видов и количество сохраняемых изолятов, представлена информация о формах хранения, описан технологический регламент обслуживания и пополнения хранилища ангидробиозных культур. Хранилище находится на стадии формирования. Его будущее связано с расширением фонда за счёт морских, пресноводных и галобных видов. Оптимизация способа обезвоживания позволит перевести в состояние ангидробиоза микроводоросли, относящиеся к разным систематическим отделам.
dc.description: Reliable preservation of microalgae cultures and creation of genetic banks of strains is one of the important tasks of modern biology. To date, 792 collections of various cultivated organisms from 76 countries are registered in the catalog of the World Federation for Culture Collections in the WDCM CCINFO database. This is the most extensive consolidated database of culture collections, which includes both well-known large collections and small repositories of research and educational institutions from all over the world. The database contains 47 algological collections and 80 collections of various microorganisms, which also include microalgae and cyanobacteria cultures. Only 30 biological collections are registered in Russia, from which only 13 contain algae strains. The most common technique of microalgae cultures storage is the method of their periodic re-sowing onto liquid media or agar. It is used in 127 collections (99 % of the total number in the catalog). Other methods used are: cryopreservation – in 33 collections (27 %), lyophilization – in 13 (11 %), L-drying – in 5 (4 %), freezing – in 19 (16 %), and immobilization in alginate beads – in 1 (0.8 %). However, when using these methods, there is a change in morphological and functional features of cells of the cultures stored, as well as their shredding. In addition, cultures maintaining in a viable state is time-consuming and requires expensive equipment. Preservation of microalgae, transferred to the state of anhydrobiosis by dehydration, is simple and cost-effective. Anhydrobiosis is a deep and long-term inhibition of metabolism, reversible under favorable conditions; it is a quite common phenomenon in nature. The only collection in the WDCM CCINFO database that applies the method of transferring cells to a resting state (for soil algae) is the collection of algae cultures of the National University of Kyiv (ACKU WDCM 994). Many years of experiments on the transfer of microalgae to the state of anhydrobiosis allowed us to develop a method of long-term preservation of microalgae without the use of nutrient media. This technique includes cells transfer to the state of anhydrobiosis, their preservation in a dehydrated state, and subsequent removal to an active culture. In order to preserve algological biodiversity, IBSS RAS created a repository of microalgae transferred to the state of anhydrobiosis, which can be converted to active cultures if necessary. The objects of the repository were marine unicellular algae, as well as freshwater and halobic species of lower phototrophs which are perspective for biotechnology and aquaculture. The cultures were obtained as an inoculum from IBSS RAS collection of live cultures of planktonic microalgae. The algae were grown in an accumulative mode under constant lighting. The biomass was collected during cultivation of algologically pure microalgae cultures at the growth retardation or at the stationary stage. Cells were separated from the culture medium by centrifugation or by filtering them on a plankton sieve. Then the algae were dehydrated and maintained in hermetic zipper bags placed in plastic containers of 100 to 500 ml, at a temperature of +18...+21 °C in the dark in a specially equipped room. The main part of the collection is represented by strains from the phyla Chlorophyta, Cyanophyta, Bacillariophyta, and Rodophyta. The list of species, the number of isolates stored, and the information on preservation forms are provided in this article. The technological regulations for maintenance and replenishment of the storage of anhydrobiotic cultures are described. The repository is at the stage of formation. Its future lies in the fund expansion to include marine, freshwater, and halobic species. Optimization of the dehydration method will allow the transfer of microalgae belonging to different systematic phyla to the state of anhydrobiosis.

dc.description.abstract: Несмотря на исключительную важность диатомовых в природных экосистемах, их генетический и биопродукционный потенциал почти не используется вследствие слабой изученности биологии и чрезвычайно низкой представленности в коллекциях культур. Отсутствует необходимый опыт выделения клонов (штаммов) диатомовых, постановки и содержания в культурах, недостаточно полны представления о жизненных циклах диатомовых. На Карадагской научной станции (Крым) занимаются изучением диатомовых водорослей более 40 лет. К настоящему времени сформирована рабочая коллекция, которая включает более 400 клонов, относящихся к 27 морским и 7 пресноводным видам. Регулярно пересеваемые культуры содержатся на жидких средах. Помимо Чёрного моря, морские виды отобраны в различных регионах всех океанов, а пресноводные – в отдельных регионах Евразии. Результаты исследования коллекции культур необходимы как для решения ряда научных проблем (выявления репродуктивных особенностей, биогеографии видов, внутривидовой изменчивости), так и для потенциального практического использования диатомовых водорослей в биотехнологиях.
dc.description: Diatoms, the essential member of the global biosphere, are represented by 30 000 known species and presumably can include about 100 000 species. These microalgae have evolved unique set of genes, pathways of biosynthesis, and physiological properties. This rich genetic and production potential is inadequately used because the biology and life cycles of these microalgae are vaguely known, their presence as living organisms in culture collections of any country is very limited and the experience in diatom clone/strain isolation and culture maintenance — in deficiency. Formation of a collection of diatom cultures is the first step to practical uses of the rich genetic and metabolic material. The sensitivity of diatoms to environmental/medium quality and to metabolites (autoinhibition) makes the task uneasy. At Karadag Scientific Station, Crimea for more than four decades the studies have been performed and the experience in clonal culture isolation and maintenance accumulated. The resulting working collection includes over 400 clones of 27 marine and 7 freshwater species. Samples of marine diatoms were gathered in the Black Sea and over the World Ocean, and the fresh-water microalgae — over Eurasian regions. The cultures are kept in liquid media and regularly reinoculated. Work with the collection is required when studying reproduction characteristics, species biogeography and intraspecific variations in diatoms, and can be of interest for biotechnologies exploring practical application of diatoms. The diatom collection at Karadag Scientific Station, its value and applications are discussed and compared with some world-known microalgal collections.

dc.description.abstract: Биологические коллекции –– основа для таксономических исследований и изучения биологического разнообразия регионов. Задача настоящей работы –– создание электронного каталога коллекционных образцов и таксономической базы данных (БД), а также развитие веб-ресурса для доступа к ним с целью интегрирования биологических коллекций в исследовательский процесс. Коллекция паразитов рыб Мирового океана ИМБИ РАН послужила материалом для создания сайта, который написан на PHP с использованием сценариев JavaScript и работает под управлением веб-сервера Apache. Управление БД осуществлено при помощи СУБД MySQL. Сайт marineparasites.org включает каталог коллекционных материалов, таксономическую базу данных и библиотеку публикаций о видах паразитов рыб. Интерфейс сайта обеспечивает доступ к перечисленным ресурсам. В основе функционирования сайта лежат четыре связанные друг с другом сущности: объект хранения, организмы, таксоны и публикации. Каждому понятию соответствует таблица базы данных. В процессе ввода система автоматически формирует связи между коллекционными экземплярами организмов, их типовым статусом и прочими атрибутами. Посетители сайта при помощи выборок и фильтров делают запросы и получают целостное представление о коллекции. Страница таксона состоит из данных о систематическом положении вида и его синонимах. На странице о виде-паразите расположен индикатор наличия экземпляров вида в коллекции, перечень атрибутов коллекционных экземпляров, публикации, где упоминается данный вид, типовой район и хозяин вида-паразита, а также другие места и хозяева находок, описания и изображения видов. Таким образом, пользователь планомерно переходит от описаний видов паразитов к их хозяевам, от коллекционных образцов –– к изображениям и диагнозам видов, к публикациям и обратно. Сайт сделан англоязычным, что даёт возможность зарубежным и отечественным исследователям получить информацию о коллекции паразитов рыб Мирового океана ИМБИ РАН и интегрировать её в международные веб-ресурсы по биоразнообразию. Аудитория сайта –– профессиональные исследователи паразитов, но заложенные идеи позволяют достроить сайт до виртуальной коллекции любых организмов или до веб-портала о разнообразии флоры и фауны любого региона. Сайт является результатом плодотворного взаимодействия биологов и программистов, способствует интенсификации обмена коллекционными данными и образцами и служит инструментом исследования паразитарной компоненты морских сообществ.

The aims of this work are the creation of an electronic catalogue of collection specimens and taxonomic database (DB), as well as the development of a web resource to access them, for integrating biological collections in the research process. The IMBR Collection of Marine Parasites is used as material for this site. The PHP and JavaScript programming languages are used for its creation and the relation database management system MYSQL is used for manage database. The website and database are operated by the Apache webserver. Website marineparasites.org includes the electronic catalog of the collection, the taxonomic database and the repository of publications about species of fish parasites. The website provides an interactive interface to access the above listed resources. The functioning of the website is based on four entities related to each other: a collection object, specimens, taxa, and publication. Each entity corresponds to a database table. During filling the tables, the system automatically creates connections between collection samples, their type and taxonomic status and other attributes. Visitors of the site can make requests using filters and get a complete representation on the collection. Webpage of taxon consists of the data on the species systematic position and its synonyms. Indicator of the presence of samples of the species in the collection, a list of attributes of collection exemplars, publications that mention this species, type locality and host of the parasite as well as other regions and hosts of its finding, description and images are located on the page of the parasite species. Thus, the user follows from the descriptions of the parasite species to their hosts, from the collection samples to their images and taxonomic diagnosis, publications on this parasite and inversely. English language of interface facilitates that more researchers can obtain information on the collection of marine parasites maintained by Institute of Marine Biological Research RAS and helps to integrate it into the international web resources on biodiversity. The site reports specific information to professional researchers of parasites, but ideas it is based on allow to develop it into a resource on collections of any organisms and diversity of flora and fauna of any regions. The site is the result of fruitful cooperation between biologists and programmers; it promotes the intensification of exchange of collection data and samples, and is a tool for studying parasite components of marine communities.
